This randomized, double-blind, placebo-controlled trial examined nicotinamide riboside (NR) supplementation in 24 healthy middle-aged and older adults. Participants received escalating doses (250mg, 500mg, 1000mg daily) or placebo over 8 weeks.

Results demonstrated that NR supplementation safely and effectively increased NAD+ levels in humans. NAD+ increased in a dose-dependent manner, with the 1000mg dose producing the most robust effects. The supplement was very well-tolerated with no serious adverse events.

NAD+ is a critical coenzyme involved in energy metabolism, DNA repair, and cellular health. NAD+ levels decline with age, and this decline is associated with age-related cognitive impairment and neurodegeneration. By restoring NAD+ levels, NR may support mitochondrial function, enhance neuroprotection, and promote healthy brain aging. This study provides important evidence that oral NR supplementation can effectively boost NAD+ in humans.
